Making a rhythm game level

I've been thinking about making a rhythm game-like level along the lines of a great LBP2 level called Pianomania, which you can see a demonstration of here.

I can probably figure out control input settings with some practice (I'm hoping to incorporate both buttons and touch controls), but am mainly figuring out how to properly time the symbols that correspond with the music. I'm guessing I should use a sequencer, but is there a good way of timing everything, or will it likely require a lot of trial and error?

Another little touch I'd like to add that will probably require less frustration is the implementation of a combo-tracking meter for fun, complete with text pop-ups every 50 or so notes a la Guitar Hero 3. Is that possible?
